Common speculation:

Just a simple camera flash problem. (Doesn't seem too likely.)

A shoulder button for sideways-controller play. (It would be in the right spot for right index finger.)

A volume control for the speaker.

Perhaps a sensativity control for the controller itself? It's far-fetched, but it seems better to have that option on the actual controller than having to change it for each game in the options menu.

Or, most likely, it's just an ID sticker for the dev kits.

To me, it looks like a peice of blue tape. That really crappy kind that never stuck to anything but your high school forced you to use it to hang posters because they got a cut of profits from the only store in town that sold it. Wait, what are we talking about again?

If anything, it's a backlight button. This way, you can play games in the dark. I have a button on the same placement on my LG TV remote.

Yeah, it's definately a dev-only thing. It's a bit darker than the Wiimote's color scheme, and the controller is dev-looking to begin with.

I say it adjusts the sensitivity or something.

it could be a springloaded button that is used to clip the wiimote in the zapgun attachment.

Oh, you mean the backlight for all the buttons. haha, nevermind. I thought you meant there was some sort of LCD on the controller or something.

In a IGN.com article they stated about a Syncho button on the inside of the battery comprtment, maybe its there!! or maybe thats ust a developer old REVOLUTION wii-mote.

maybe a walkie-talkie-push-to-talkie-button.. or just a piece of plastic that covers up another mysterious slot.. hopefully a button to give the other players an electric shock.. or a little sponge to dry your sweaty thumb
however if it really turns out to be a button it will be for something u hardly ever use ingame (just to say something clever.. obvious.. but clever)

Didn't they say that both the console and the controller would have a synchro button? I think this is it.

It doesnt look at all in the right place for a classic-style shoulder button. It would be on the bottom of the pad, not the top.
maybe: Sync/console power button.
On-Off switch for the rumble motor.

The controller's sync button is inside the battery compartment.

"Evidently gamers first press the SYNCHRO button on the console itself and then find and press another SYNCHRO button located inside the battery compartment of the Wii controller. Documentation also suggests that gamers can hold down the 1 and 2 buttons on the Wii-mote to accomplish the same task."

NO it's either a volume control for Wii's speaker or
a standard button that you can use like for an extra gadget or somethin' 'cause Miyamoto San said they were lookin' for more options for the controller...Or
it's a release button for the other attachments, like the nunchaku or the zapper...Or
it's some kind'o button for the classic style (the one you turn around to play NES games) that you can use.

The sync button is inside the controller (where you put the batteries).

That button isn't on any other Wiimote in that magazine.

It's just a glare or tape. Nothing more, nothing less.

The colors were wrong, the controller was evidently hardwired (look closely at the protruding wire, the controller is wired to a dev kit probably), and that makes it seem like a dev kit exclusive button, possibly for time travel, but probably something more mundane, like lasers. Who knows though. Could be something cool.

I think I noticed another secret button. Look by the left side of the giant red arrow. A green discoloration should be apparent. Color problem, or new, unknown button? And that woman! She has grainy face syndrome!

It looks like a Gamecube "Z" button to me.
