1) Gesture Jams
The game's signature moves are obviously one of its main attractions, and adding some motion control for the Jams could easily provide for some hilarious, over-the-top moments. The better the animation, the more fun Gesture Jams would be.
2) Character creation
Instead of scoring crazy 3-pointers and knocking other players flat on their backs with your everyday cast of famous, overpaid athletes, why not allow for character creation? The incredibly lanky, the comically rotund -- perfect for an outrageous basketball game. Even better, a create-a-character would also expand upon Jam's existing stat system, as every player had unique attributes. I can already envision a character who dunks like a maniac but can't make a 3-pointer to save his life.

3) Tag moves
The two-on-two system is practically begging for some cooperative tag dunks and crazy scores. Imagine launching your teammate across the court like a human torpedo, basketball in hand.
4) Trick points
NBA Street is likely the only currently-running series to share much in common with NBA Jam, and the game's trick system could work well in tandem with the Jams. Ball-stealing, evasions, and flashy moves could go into a point total to be used to unlock things in-game, flesh out the create-a-character, or affect the outcome of a match.
