A few days after voice actor Jon St. John (best known as Duke Nukem) tweeted about doing work on Valve's unannounced Defense of the Ancients, Superannuation has noted a trademark filed by the publisher with the US Patent and Trademark Office for "DOTA."
We're following up with Valve again for an official response, but you know how this one goes. It's okay, Valve -- you can talk to us. There's no shame in working on DotA. We're okay with it, nobody is judging you -- we'll still give you bro hugs and use Steam even after you come out with it.

Valve has 200+ employees. Their projects typically consist of around 30-40 people, more if it's something xbox hueg like Half-Life 2. The team being led by IceFrog is different than the team working on the next Half-Life game.

It's almost like they're TRYING not to show us Episode 3... Everything imaginable is being released by Valve EXCEPT the next Half-Life game. I'm happy with that (kind of), Portal 2 looks awesome and that Alien Swarm games pretty cool... But seriously, they know we want it, WE know they're working on it... Just give us a tidbit, ANYTHING!

Now why would they do that? Then you would know that is officially coming, lol it be awesome just to come home from work/school and see a commercial saying Half-Life 3 was coming out the very next day.

I hope this is going to be an all new-standalone game!
I loved DOTA but I don't have WC3 anymore. Hopefully this game will have nice graphics and etc. etc. etc. and of course not too expensive!

DotA is a game where there are two more-or-less identical teams on the opposite sides of a valley. Each team has a number of players that are heroes, units that can level up over time and equip items. Each hero also has a series of unique abilities. Each team also has lanes of combat where automated units are constantly spawned through at the same pace for each side. There are various chokepoints in the lanes that have towers controlled by the AI. These towers attack units and heroes of the opposing team. The ultimate goal of the game is to reach the other side's ancient, which is in the center of a large base where units are spawned from and where they split off into the lanes. Killing the ancient wins the game.
