From what I've read, since the events of the first game, Santa Destroy has become more of a tourist spot so it'll be a lot more populated with more to do. At least that's what I got from the Famitsu article.

E3 is obvious, 5 COULD mean MGS5 but it could be too soon, and whatever that last symbol is (we could assume it's a lowercase 'e'), who knows what that means.

In the Digest option, you CANNOT save your progress, meaning you can't start off of a later point in the game and progress from there with all the gear you may have collected in that period of time. You can only do that in the regular game mode.

It's also possible that this means the difficulty COULD be ramped up a bit, or preferably, different difficulty settings.

I would launch Rok-It Launchers from my Rok-It Launcher, which in turn would be launching other Rok-It Launchers. Try to wrap your mind around that one.
